PhD position: Propagation of uncultivable EV/HPeV infection in human 3D models

What you are going to do
This research project focuses on researching the propagation of uncultivable EV/HPeV infection in human 3D models. During this project, you will be focussing on the following research objectives:
To culture EV-C types 104, 105, 109 and 116-118 positive samples on 3D models
To culture HPeV8-16 positive samples on 3D models
To study virus replication kinetics, viral entry, and effects on gut, lung/HAE or brain organoids
With this project, we aim to achieve the following results:
The successful propagation of one or more EV/HPeV for further characterization of viral replication, tropism and virus-host interaction.
This project also includes two secondments, which are as follows:
A seven-month secondment with The National Institute for Public Health and the Environment (RIVM) focused on bringing HAE to the RIVM and to collect samples for cultivation (location: Bilthoven, the Netherlands)
A three-month secondment with the University of Clermont Auvergne focused on learning about the mini-brain model (location: Clermont-Ferrand, France)
You will be guided during your research project by Katja Wolthers (Department of Medical Microbiology) and Dasja Pajkrt (Paediatric Infectious Disease Specialist).
This research project is provided by Organovir, an Innovative Training Network (ITN) that focuses on the implications of organoid technology for viral pathogenesis studies and anti-viral testing. Organovir has an international consortium that exists out of 10 beneficiaries and 5 partners from various sectors, such as academics, industry as well as the governmental and non-profit sector. One of these beneficiaries is the Amsterdam UMC.
As a researcher with Organovir, you will be able to familiarize yourself not only with the scientific and technical aspects of innovative 3D culture systems for virus research, but also with the financial, commercial, ethical, communicative, policy and dissemination aspects enabling optimal valorisation of organoids. At the end of this project, you will hold a unique position within the European labour market

What we expect from you
For this research project we are looking for a young researcher who is highly motivated and enthusiastic with a flexible, pro-active team spirit and a recent Master degree in the field of biochemistry, cell biology, molecular biology, virology, tissue engineering or life sciences. Interest and experience in virus culture and organoid technology is an advantage. The candidate is required to be proficient in the English language and should be in the first four years (full-time equivalent) of their research careers. In addition, the applicant should not, at the time that he/she is selected by the host institution, have resided or carried out their work and/or studies in the country of the host institution for more than 12 months in the 3 years immediately prior to their recruitment. What we offer you
We offer a 4-year research position for 36 hours per week within a motivated and innovative consortium. The salary is in accordance with CAO-UMC, scale 21. In addition, we offer a holiday allowance of 8% of the gross annual salary. The starting date of the research project is the 1st of September 2019.
